چکیده

The in situ uptake from the rat jejunum of liposomes prepared fr >m various saturated phosphatidylcholines in the presence or absence of cholesteol was determined. Liposomes prepared from distearoylphosphatidylcholine. with or without cholesterol, were taken up at the fastest rate (apparent first-order uptake of intact liposomes with a half-life of about 15 min). The lipid components OE liposomes could not be found in compartments where liposomes would be e’vpected to accumulate, i.e. liver, spleen. lymph nodes, or thoracic lymphatic duct. Ftuthermore. inulin, when encapsulated in the liposomes, could not be found in the urine. When either PEG-4000 (a non-absorbable marker) or glucose (an absorbable marker) was entrapped within the liposome, all of the liposomes and markers that we.e taken up still remained associated with the jejunum and the liposomes remained ir,ltact.

Barriers to liposomal gene delivery: from application site to the target

AbstractGene therapy is a therapeutic approach to deliver genetic material into cells to alter their function in entire organism. One promising form of gene delivery system (DDS) is liposomes. The success of liposome-mediated gene delivery is a multifactorial issue and well-designed liposomal systems might lead to optimized gene transfection particularly in vivo. Liposomal gene delivery systems...

Gastrointestinal uptake of liposomes. II. In vivo studies

The effect of liposomal entrapment on oral absorption of drugs was deter~ned from the excretion profiles of entrapped markers after oral administration to rats. Liposomal entrapment does not facilitate the uptake of PEG-4000, a non-absorbable, water-soluble marker. The excretion patterns of hydrocortisone and salicylic acid, when entrapped in L-a-phosphatidylcholine distearoyl/cholesterol or eg...

Thermoreversible Pluronic® F127-based hydrogel containing liposomes for the controlled delivery of paclitaxel: in vitro drug release, cell cytotoxicity, and uptake studies

PURPOSE To develop an in situ gel system comprising liposome-containing paclitaxel (PTX) dispersed within the thermoreversible gel (Pluronic® F127 gel) for controlled release and improved antitumor drug efficiency. METHODS The dialysis membrane and membrane-less diffusion method were used to investigate the in vitro drug release behavior. Differential scanning calorimetry (DSC) thermal analys...

The intestinal uptake of "enzymatically-stable" peptide drugs in rats as influenced by D-glucose in situ.

In previous in situ and in vivo rat perfusion studies, the intestinal absorption of several low molecular weight drugs was increased by the presence of luminal D-glucose. The intent of this study was to determine the potential of this fed-state effect to improve the intestinal uptake of poorly permeable, small peptide and peptide-like drugs. Jejunal wall permeabilities (Pw*) of di-(D-kyotorphin...
